Quick Computer Hardware question

Aite, so I'm purchasing an eSATA to SATA cable so I can copy stuff from my broken computer to my laptop (that has an eSATA port). Now if I was to turn the PC power on and have the e/SATA cable plugged into the different hard drives, would it be cool just to plug in/out of the hard drives while they're on or will I have to turn the PC on and off each time?
