Enoch, a 19-year-old who also plays as a forward, managed 80 minutes of action before being substituted with cramp.
Like Mario, who was on the bench as City's Champions League campaign came to an end on Wednesday, Enoch was born in Ghana but raised in Italy.
The younger sibling has previously caught the eye of Serie A side Napoli - the team who, ironically, knocked the Premier League leaders out of Europe - but is understood to have been training with Stoke for three weeks.
